Factor V Leiden is the most common inherited thrombophilia (blood clotting disorder), affecting approximately 5% of Caucasians. The mutation makes Factor V resistant to inactivation by Activated Protein C, leading to increased clot formation. Heterozygous carriers (one copy) have a 3–8x increased risk of venous thromboembolism (VTE), while homozygous carriers (two copies) have a 50–80x increased risk. This test is recommended for individuals with a personal or family history of blood clots (DVT, pulmonary embolism), unexplained recurrent pregnancy loss, or clots at a young age. Results inform decisions about anticoagulation therapy, hormone use, and surgical precautions.
